Version Fran?aise English versionUn dialogue permanent s¡¯¨¦tablit dans le tube digestif entre le micobiote et l¡¯h?te et tout d¨¦s¨¦quilibre de cette symbiose est ¨¤ l¡¯origine de nombreuses pathologies. L¡¯UMR 1071 Inserm/Universit¨¦ Clermont Auvergne m¨¨ne depuis vingt-cinq ans les travaux consistant ¨¤ analyser la relation bact¨¦ries-h?te en ¨¦tudiant les Escherichia coli pathobionts impliqu¨¦s dans les maladies inflammatoires chroniques de l'instestin, en particulier la maladie de Crohn (MC). Nous avons ¨¦t¨¦ pionniers dans la mise en ¨¦vidence que la muqueuse il¨¦ale des patients atteints de la MC est anormalement colonis¨¦e par les souches pathog¨¨nes de Escherichia coli adh¨¦rents et invasifs (adhrerent-invasive E. coli ou AIEC) qui sont capables d¡¯adh¨¦rer et d'envahir les cellules ¨¦pith¨¦liales intestinales, de survivre et de se multiplier dans les macrophages, induisant une r¨¦ponse pro-inflammatoire. Les bact¨¦ries AIEC sont ¨¦galement capables de persister dans le tube digestif, de coloniser la muqueuse intestinale et d'induire l'inflammation chez les souris g¨¦n¨¦tiquement pr¨¦dispos¨¦es. De plus, en raison du lien fort qui existe entre inflammation chronique et cancer, nous avons ¨¦galement ouvert un nouveau sujet d¡¯¨¦tude concernant la piste infectieuse ¨¤ E. coli dans le d¨¦veloppement de cancer colorectal.
L'Unit¨¦ M2ISH est organis¨¦e en mono-¨¦quipe et s'articule autour de trois axes de recherche interactifs : (i) exploration de la virulence et la r¨¦sistance aux antibiotiques des Escherichia coli pathobionts impliqu¨¦s dans des pathologies chroniques intestinales, telles que la maladie de Crohn et le cancer colorectal, (ii) ¨¦tude de la susceptibilit¨¦ de l¡¯h?te ¨¤ l¡¯infection par ces E. coli pathobionts, et (iii) d¨¦veloppement de nouveaux biomarqueurs et de th¨¦rapies personnalis¨¦es innovantes
There is an ongoing dialogue in the digestive tract between microbiota and the host. Any imbalance in this symbiosis leads to many diseases, such as chronic inflammatory bowel diseases, colorectal cancer, obesity, metabolic syndrome. For the last ten years, the research unit ¡°Microbes, Intestine, Inflammation and Host Susceptibility¡±, UMR 1071 Inserm/Universit¨¦ d¡¯Auvergne, INRA USC 2018, has been studying the relationship between intestinal microbiota and the host, particularly the involvement of pathogenic Escherichia coli in chronic inflammatory bowel diseases, focusing on Crohn's disease and the role of enterohemmoragic E. coli involved in acute diarrhea and hemolytic uremic syndrome. This research unit is pioneer in demonstrating that the ileal mucosa of patients with Crohn's disease was abnormally colonized by pathogenic adherent-invasive Escherichia coli due to genetic susceptibility of Crohn¡¯s disease patients with increased expression of intestinal receptor allowing E. coli colonization and/or with defects in autophagy, a cellular process allowing host cells to eliminate any intracellular pathogens. In addition, due to the strong link between chronic inflammation and cancer, this research unit also studies the link between modifications of the intestinal microbiota and colorectal cancer.
This unit is currently developing three areas of research: (i) exploration of the virulence of Escherichia coli responsible for chronic inflammatory bowel diseases and colorectal cancer, (ii) analysis of the host susceptibility to develop Crohn's disease or colorectal cancer, and (iii) design of new specific therapies.
